Blasticidin S hydrochloride 10 mg

Blasticidin S is a peptidyl nucleoside antibiotic isolated from Streptomyces griseochromogenes. It specifically inhibits protein synthesis in both prokaryotes and eukaryotes by interfering with the peptide bound formation in the ribosomal machinery. Resistance to blasticidin is conferred by the blasticidin resistance gene from Bacillus cereus (bsr) which codes for blasticidin deaminase. Typically, bacteria are sensitive to blasticidin concentrations of 25-100µg/mL, and mammalian cells to 1-10µg/mL. In the mean-time different Blasticidin resistance gene have been identified: one acetyltransferase [bls] and two deaminases [bsr and BSD]. These genes are used as genetic marker. As the possible working concentration for Blasticidin is very broad and differs from organism to organism respective from cell line to cell line, it is highly recommended to determine the needed value. High salt concentrations inhibit Blasticidin S. Therefore the sodium chloride concentration has to be reduced from 10g/mL to 5g/ml for the screening of Blasticidin S resistant E.coli. Stability: Powder is stable for 2 years at 4°C. Wie recommend to sterile filter all Blasticidin solutions. Stock solution of 50mg/mL Lösung should be used. The pH-value of the solution must be below or at pH7, as Blasticidin is inactivated at pH-values higher than 7.0. Solution containing Blasticidin are stable for 2 weeks at 4°C, or 2 months at -20°C. Repeated freeze-thaw cycles are not recommended.
