Mainly used for staining nucleic acids in polyacrylamide gels, or to stain agarose gels.

Methylenblue is used to stain nucleic acids in polyacrylamide gels, but it can also be used to stain agarose gels. Staining procedure: Incubate gel for 1 hour in 0.4 M Na-acetate/0.4 M acetic acid / 0.2% methylenblue (pH4.7). Peacook A.C., Dingman C.W. (1967) Biochemistry 6, 1818-1827.

Application: Used to stain RNA in polyacrylamide gels at pH below 6.0.
