Proteins, RNA and DNA molecules are denatured by Urea. Used in polyacrylamide gels by adding 7M urea. (Maniatis T., Efstratiadis A. (1980) Methods Enzymol., 65, 299). Nucleic acid molecules larger than 150-200 nt, do not denature completely at room temperature in 7M urea. At alkaline pH, urea decomposes and forms cyanate ions. These ions may react with the amino group and form stable carbamylated derivatives that may result in a change of the migration pattern during electrophoresis. solutions that contain urea should not be stored for long periods or be heated. Gels containing urea should be prerun to remove cyanate ions before loading samples.
