b-Agarase is used for the recovery of pure DNA and RNA from agarase low melting gels (Ishumatsu K. et al. (1956) Sci. and Ind. (Japan), 30, 137). One b-Agarase unit will solubilise 100 mg of low melting agarose gel (1%) at 40-42°C within 30 minutes. To isolate nucleic acids from the agarose gel, cut out the band from the gel as accurately as possible; add to the slice half of the volume (or weight) of 1X reaction buffer and melt the agarose at 40-42°C. Add one unit b-Agarase per 100 µL melted agarose and allow to incubate at 40-42°C for one hour. Nucleic acids can then be recovered by a simple ethanol precipitation. |
