Construction of recombinant Baculo virus clone

2 mL each of rec. virus supernatant as viral stock, plus 1 mL

Co-transfection of Sf9 insect cells with recombinant transfer vector, followed by isolation and plaque purification of 10 recombinant Baculo-viruses from supernatant. Cell infection with each recombinant and preparation of viral DNA. DNA-samples will be analysed to make sure they contain gene of interest. 5 of the positive rec. viruses are used to infect Sf9 cells (4x10(6)).
