Highly purified modified Arginine-C endopeptidase with extremely low autolysis and therefore greater control of protein fragmentation. Consistent Enzyme activity over long digestion periods. Characteristics: Arginine-C endopeptidase (Clostripain) is a sulfhydral endopeptidase, which specifically hydrolyzes the carboxy peptide bond of Arginine. It is produced in cultures of the bacterium, Clostridium histolyticum, as an extra cellular enzyme. As a sulfhydral enzyme Arginine-C is susceptible to inactivation by oxidation and it requires reducing agents for protection. The enzyme also requires calcium ion for maximal activity. A special reconstitution buffer is supplied which contains reducing agents and activators to maintain enzyme activity. Chemical Modification: Highly purified enzyme is modified chemically by a special process. As a result, the modified enzyme is more resistant to autolysis and has improved stability. Thus the modified enzyme retains at least 80% of its activity after 6 hours incubation at 30°C in reaction buffer and at least 65% of its activity after 24 hours incubation under the same conditions, while retaining its specificity for Arginine.
