Highly purified modified Chymotrypsin with extremely low autolysis and therefore greater control of protein fragmentation. Consistent Enzyme activity over long digestion periods. Characteristics: Chymotrypsin is a serine endopeptidase which predominantly cleaves peptide bonds on the carboxy side of Tyrosine, Phenylalanine and Tryptophan. In addition, Chymotrypsin also hydrolyzes the carboxy side of Leucine, Methionine, Alanine, Aspartic and Glutamic acids, althoµgh at a much lower rate. This highly pure, sequencing grade, modified Chymotrypsin is first treated with TPCK and then subjected to an extensive purification process to remove contaminating protease and Chymotryptic autolysis by-products, which could affect the specificity of the digestion process. Subsequently, the enzyme is chemically modified to minimize autolysis and to increase stability. Applications: Modified Chymotrypsin is supplied lyophilized in 25 µg vials. The enzyme is typically reconstituted to a concentration of 1 µg/µL with deionized H2O or buffer of choice (25µL per vial). The modified enzyme retains in excess of 80% of its activity after six hours at 30°C in reaction buffer, and in excess of 70% of its activity under the same conditions after 24 hours.
