Bacterial Alkaline Phosphatase (BAP)

The enzyme catalyses the removal of 5'-phosphate groups from DNA, RNA and ribo-or deoxy-ribonucleoside triphophates.

For efficient cloning experiments, dephosphorylation or phosphorylation of blunt or overhanging 5' ends is essential. For dephosphorylation, the Bacterial Alkaline Phosphatase (BAP) is used. For phosphorylation of oligonucleotides and dephosphorylated DNA or replacement of existing 5'-phosphate with gamma-PO4, T4 Polynucleotide Kinase (PNK) is used.

Unit-definition: One unit is defined as the amount of enzyme which hydrolyses 1 µmole of 4-nitrophyenylphosphate in 1 minute at 37°C.
