For efficient cloning experiments, dephosphorylation or phosphorylation of blunt or overhanging 5' ends is essential. For dephosphorylation, the Bacterial Alkaline Phosphatase (BAP) is used. For phosphorylation of oligonucleotides and dephosphorylated DNA or replacement of existing 5'-phosphate with gamma-PO4, T4 Polynucleotide Kinase (PNK) is used. |
