Albomycin is an antibiotic, that is structurally related to the hydroxamate siderophore Ferrichrome. Albomycin is activally taken up from E.coli by the Ferrichrom transporter system, consisting of the outer membrane proteine FhuA, the ABC transporter FhuBCD and the energy transfer complex TonB and ExBD. This uptake needs energy. Within the cell Albomycin is cleaved by the peptidase N. After cleavage the antibiotic active part (aminoacyl thioribosyl pyrimidine) is found within the cell, while the iron carrying part is secreted into the medium. The thioribosyl-pyrimidine derivative inhibits (in vitro) Seryl-tRNA-Synthetase. In Salmonella thyphimurium both Peptidase N and A are active in cleaving Albomycin. E.coli Peptidase N mutants are able to take up Albomycin, but are unable to cleave the substance. This way the cell can use Albomycin as a Siderophor (Ferrichrom analogon) for their iron supply. |
