MGMT (Human) ist auf dem Chromosome 10 (10q26) lokalisiert. Es kodiert für das DNA-Reparaturprotein: O-Methylguanine-DNA Methyltransferase. MGNT repariert alkylierte Guanidine in DNA durch stöchiometrischen Transfer von Alkylgruppen an der O-6 Position auf ein Cysteinrest im Enzym. Dabei kommt es häufig zum Verlust der Genexpression durch Hypermethylierung der CpG sites innerhalb der Promotorregion des MGMT-Gens.

Assay Data
The region of interest is within the promoter region of the MGMT gene, 27 CpG sites are observed in this ROI.
Therefore the assay contains 2 parts, whether you choose one or both belonging to your interest.

Analyzing Method
1) Genomic DNA is treated with bisulfite
2) Amplification of the region of interest
3) Methylation level of each CpG site is determined by inhouse Pyrosequencing®-Technology
4) Quality Control of bisulfite rxn and sequence context are included.
