Dithioerythritol (DTE) ist ein Isomer des Dithiothreitol (DTT). Prinzipiell ist DTE gegen DTT austauschbar bzw. DTT gegen DTE. DTE hat pK-Werte von 9,0 und 9,9 und DTT von 8,3 und 9,5. Da beide Verbindungen nur im ionisierten Zustand funktionieren, ist ihre reduzierende Wirkung im pH-Bereich 7,0 - 9,5 am höchsten. Im niedrigeren pH-Bereich ist DTT aktiver als DTE. Die reduzierende Wirkung kann durch Ansäuern (z. B. Essigsäure auf pH 3) gestoppt werden. Proteine können während der Gelelektrophorese oxidieren. Dies resultiert in breiteren Banden (verminderte Auflösung) und nicht reproduzierbarer Mobilität (Fehleinschätzung des Molekulargewichtes). Reduktion der Proteine mit DTE (0,25 M Stammlösung in Wasser, 5 mM Endkonzentration) und Alkylierung mit Iodacetamid (0,25 M Stammlösung, 1 : 20 verdünnt in der Endkonzentration) verschafft Abhilfe.
