Ribonuklease A ist eine Endo-Ribonuklease, die spezifisch Einzelstrang-RNA am 3'-Ende von Pyrimidinbasen (Cytosin und Uracil) schneidet. RNAse A wird benutzt, um RNA-freie DNA zu erhalten und um nichthybridisierte RNA-DNA Hybride zu hydrolisieren. Das pH-Optimum von RNAse A liegt zwischen 7,0 und 7,5. RNAse A wird durch Diethylpyrocarbonate (DEPC) >, Guanidiniumsalz (4 M Guanidiniumthiocyanat), beta-Mercaptoethanol, Schwermetalle, Vanadylkomplexe, RNAse-Inhibitor inhibiert. RNAse A spaltet Einzel- und Doppelstrang RNA und RNA in RNA:DNA Komplexen bei niedriger Salzkonzentration (100 mM NaCl). Bei hohen Salzkonzentrationen (>300 mM NaCl) spaltet das Enzym nur noch Einzelstrang RNA. Das Enzym kann durch Proteinase K-Verdau und anschließende Phenolextraktion entfernt werden.

Eliminierung von potentieller DNAse-Aktivität:
Produkt S5231 (RNAse A (nicht zertifiziert DNAse-frei)) kann DNAse-Aktivität beinhalten. Da jedoch RNAse A hitzestabil ist empfehlen wir eventuelle DNAse-Aktivität durch Hitzeinaktivierung zu entfernen.

Prozedur:
10mg/mL RNAse A gelöst in 0,01 Natriumacetat (pH5.2) werden 15 Minuten lang bei 100°C for 15 im Wasserbad inkubiert. Anschließend verbleibt der Ansatz im Wasserbad, während dieses auf Raumtemperatur abkühlt. Der pH-Wert der Lösung kann durch Zugabe des 0,1-fachen Volumens an 1M Tris-HCl (pH7,4) angepasst werden. Nach dem aliquotieren, der nun DNAse-freien RNAse A, sollten alle Aliquote bei -20°C gelagert werden.
ACHTUNG: RNAse A präzipitiert wenn konzentrierte Lösungen bei neutralem pH-Wert auf 100°C erhitzt werden!

Stock solution: Stammlösungen können im Konzentrationsbereich von 1 - 10mg/mL in 10mM Tris/HCl, pH7.5; 15mM NaCl oder in 10mM Tris/HCl, pH7.5; 1mM EDTA, pH8.0 (TE-Puffer) hergestellt werden. Die empfohlene Arbeitskonzentration ist 10µg/mL (für die Entfernung von RNA aus Plasmidpräparationen; 1 hr, RT) oder 100ng/mL (bei der Präparation von 'blunt ends' bei doppelsträngiger cDNA).

Stabilität: RNase A aggregiert während der Lyophilisation und Lagerung. Das Protein hat eine hohe Affinität zu Glassoberflächen, was bei der Herstellung von Lösungen berücksichtigt werden sollte. Bei neutralem pH (z.B. in PBS pH 7.4) und hohen Konzentrationen (>10mg/mL) wird RNase A präzipitieren. Bei +2°C to +8°C (lyophilisiert) ist RNase A für mehrere Jahre stabil (Lagerung im Trockenen). In Lösung (-20°C) ebenfalls für mehrere Jahre und bei +2°C to +8°C für einige Wochen.
