Die leicht zu inaktivierende Genaxxon Proteinase Low-Temp ist eine unspezifische Endopeptidase, die aus einer arktischen marinen mikrobiellen Quelle stammt. Sie hat eine breite Substratspezifität und ist nach Gebrauch leicht zu inaktivieren. Bereits ab einer Temperatur von 60°C wird die Genaxxon Proteinase Low-Temp hitzeinaktiviert und ist somit ideal geeignet für hitzeempfindliche Proben.
Die Proteinase Low-Temp ist am aktivsten im Temperaturbereich von 25°C-40°C. In der Regel wird eine Hitzeinaktivierung bis zu 15 Minuten bei 40°C-65°C empfohlen, abhängig vom Grad der erforderlichen Inaktivierung.
Es ist bekannt, dass Histone und andere Proteine Nukleinsäuren vor einer optimalen Wechselwirkung mit anderen DNA-bindenden Proteinen und Enzymen abschirmen. Die Genaxxon Proteinase Low-Temp eignet sich ideal für die Transformation von Chromatin und anderen kompakten Nukleinsäuren in bloße DNA. Das Enzym ist leicht inaktivierbar. Dies ermöglicht eine thermische Inaktivierung bei Temperaturen, die für RNA-Integrität sorgen und die Dissoziation von dsDNA vermeiden.

Anwendungsbeispiele:
- Präparation chromosomaler DNA für die PFGE
- Abbau von Nukleasen (DNase, RNase) bei der Präparation von Nukleinsäuren, einschließlich der Gewinnung nativer RNA
- Isolierung genomischer DNA aus Gewebe von Mausschwänzen
- Isolierung genomischer DNA aus Zellkulturen

Spezifische Aktivität: >30 U/mg. Fremdaktivität: RNase und DNase nicht nachweisbar. Temperatur-Optimum: +25°C bis +40°C (pH 7-10).
Empfohlene Inaktivierungsbedingungen: In der Regel bis zu 15 Minuten bei 40-65°C, abhängig vom Grad der erforderlichen Inaktivierung.
Inhibitoren: Allgemein Serinproteaseinhibitoren wie PMSF.

Anwendungshinweise: Das Enzym ist bei Lagerung von -20°C bis zu 2 Jahre lang stabil. Die Genaxxon Proteinase Low-Temp ist auch bei Lagerung von 4°C (>6 Monate) und Raumtemperatur (>4 Wochen) stabil. 
Die optimale Enzymaktivität liegt zwischen pH 7 und 10. Empfohlener Temperaturbereich für die Aktivität ist 25–40°C. Die Aktivität der Genaxxon Proteinase Low-Temp ist nicht von zweiwertigen Kationen wie Calcium abhängig (Ca2+) und ist daher in Puffern aktiv, die EDTA enthalten (<40°C Reaktionstemperatur).
Genaxxon Proteinase Low-Temp ist geeignet für den Verdauung von Proteinen in Gegenwart von SDS (0,2–1%) und Harnstoff (1–5 M). Das Enzym verträgt 500mM Guanidinthiocyanat und 1% Triton X-100 (>50% Aktivität). Es kann kein signifikanter Aktivitätsverlust bei 400 mM NaCl beobachtet werden.

The easy inactivation allows gentler inactivation compared to competing proteases. If being diligent with keeping samples on ice (e.g. RNA samples) when setting up the PCR reactions, it might also be possible to avoid the inactivation step altogether. Suitable for the digestion of proteins in the presence of SDS (0.2-1%) and urea (1-5 M). The enzyme tolerates 500mM guanidine thiocyanate and 1% Triton X-100 (>50% activity). No significant loss of activity can be observed at 400 mM NaCl.

This enzyme differs from normal Proteinase K as it is easier to inactivate and gentler on downstream procedures. Its main purpose is to reduce chromatin to naked DNA prior to analysis. It is best fitted to analysis of small amounts of cells, where endogenous proteinases will not have an adverse effect on downstream analysis

Possible protocol for tissue cells:
Seed small amounts of cells in 96-well plate (few thousands?)
Following day – wash and lyse cells in lysis buffer supplemented with AZ Protease (7.5 U/mL)
Transfer 2µL lysate to a 10µL qPCR reaction kept on ice
Run qPCR directly

Heat inactivate might be necessary in some cases (e.g. in case of not working on ice, as this means that used enzymes will be more susceptible for protease activity), then a typical inactivation step might be up to 15 minutes at 40-65°C, depending on degree of inactivation needed.

I used Proteinase Low-Temp in a saliva matrix and did not observe any amplification in the downstream qPCR analysis. What can I do?
A high concentration of Ca(2+) ions in saliva can stabilize the enzyme, making it more difficult to inactivate the Proteinase by temperature. To overcome this, add EGTA to the saliva before adding the proteinase to restore the inactivation properties.
