alpha-Chymotrypsin ist eine Serinprotease die Peptidbindungen mit aromatischen oder großen hydrophoben Seitenketten (Tyr, Trp, Phe, Leu) am Carboxyende spaltet. Dabei aktiviert und stabilisieren Ca2+ Ionen das Enzym. alpha-Chymotrypsin besteht aus 241 Aminosäuren die auf 3 Untereinheiten verteilt sind (A-Kette: 13 Aminosäuren, B-Kette: 131 Aminosäuren und C-Kette mit 97 Aminosäuren). Die Untereinheiten sind über Disulfidbrücken verbunden.

Inhibitoren von alpha-Chymotrypsin sind: Diisopropylfluorphosphat (DFP) >, Phenylmethansulfonylfluorid (PMSF) , N-p-tosyl-L-Phenylalaninchlormethylketon (TPCK) >, Chymostatin, Aprotinin, α1-Antitrypsin, α2-Macroglobulin, 10mM Cu2+ und Hg2+.
