Lipopeptide

Lipoproteine sind Teile der äußeren Membran von Gram-negativen und Gram-positiven Bakterien sowie von Mykoplasmen. Bakterielle - einschließlich mykoplasmale - Lipoproteine (bLP) sind durch den Einbau der ungewöhnlichen Aminosäure Dihydroxypropylcystein (Dhc), die durch zwei oder drei Fettsäuren acyliert ist, charakterisiert.

Synthetische Analoga (sLP) und die N-terminalen Teile des Lipoproteins von E.coli agieren in-vivo und in-vitro als potente Immunoadjuvantien. Die Immunisierung von Meerschweinchen und Schweinen mit einem komplett synthetischen Vaccin, bestehend aus dem Lipotripeptid-Adjuvant Pam3Cys-Ser-Ser sowie aus B- und T-Zell-Epitopen des Maul-und-Klauenseuchevirus, führte zu einem lang anhaltenden Schutz gegen dieses Virus.

Die Konjugation von Class-I Peptiden mit Pam3Cys-Ser-Ser war zum ersten Mal im Jahr 1989 erfolgreich und resultierte in einer effizienten Anregung von virusspezifischer T-Zell-Toxizität.

Das äußere Membran-Lipoprotein (OspA) aus Borrelia burgdorferi wurde in humanen Impfversuchen getestet und ergab sogar dann einen wirksamen Schutz, wenn es oral verabreicht wurde (Mausversuche).
