Peptidyl-Prolyl Cis/Trans-Isomerase - rHuPIN-1 - EC 5.2.1.8

Rekombinante humane Peptidyl-Prolyl Cis/Trans Isomerase. Pin 1 (EC 5.2.1.8) ist eine Peptidyl-Prolyl Cis/Trans-Isomerase (PPIase) die mit NIMA interagiert. Pin 1 enthält eine WW Proteinwechselwirkungsdomäne die essentiell für die Funktion als Zellzyklus regulierendes Enzym ist. Es ist strukturell und funktionell mit Ess1/Ptf1, einem essentiellen Protein in knospender Hefe. In Hefe konnte auch gezeigt werden, dass eine PPIase für die richtige Funktionsweise von Ess1/Pin1 notwendig ist. Somit spielt die Pin1 PPIase-Aktivität eine zentrale Rolle bei der Mitose, sehr wahrscheinlich mittels Interaktion mit NIMA.

Substrate von Pin1 sind unter anderem die Mitoseregulatoren Cdc25 Phosphatase und NIMA, PLK I, Wee, sowie Myt1 Kinase und verschiedene Transkriptionsfaktoren wie b-Catenin, c-Jun, das Tumorsuppressorprotein p53 und einige spezifische Proteine wie die RNA Pol II, das Zytoskeletprotein Tau und das G1/S Protein Cyclin D1.

Humane, rekombinante PPIase aus E.Coli isoliert ist ein nichtglykosyliertes Peptid mit 163 Aminosäuren und einer molaren Masse von 18.2 kDa.

Die Aktivität ist >330 nmol/min/mg (Assay mit suc-AAFP-pNA und Chymotrypsin bei 25°C in Tris/HCl pH8,0).
