How does it work?

Shown is exemplary genotyping data from two different blood samples.

The green melting curve originates from an EDTA blood sample and shows a single melting peak at 66 °C. This clearly indicates the presence of a homozygous wild-type genotype.
In contrast, the blue melting curve from another EDTA blood sample displays two distinct melting peaks—one at the same temperature as the green wild-type sample (66 °C), and a second at a significantly lower temperature (54 °C). The presence of two peaks is characteristic of a heterozygous genotype, indicating that two different alleles are present in the sample.
A single melting peak at the lower temperature would, in turn, indicate the presence of a homozygous mutant genotype.
Please note: minor shifts in melting temperatures can occur due to variations in salt concentration and pH in crude samples, which may influence the melting behavior slightly.

Our kits are designed to be platform-independent and system-open. They are compatible with a variety of real-time PCR cyclers. Essentially, any cycler capable of detecting fluorescence in the FAM channel and performing a melting curve analysis will work for most assays. Genotyping is determined by analyzing the melting curves. Analyze the melting peak temperature and the number of peaks. Two peaks indicate a heterozygous genotype, while a single peak suggests homozygosity. The exact genotype can be determined using the melting temperature. Please refer to the specified melting temperatures in the manual for each genotyping kit.
Our Genotyping Kits are designed for a variety of sample materials. The advantage of our kits is that you don't need to purify or extract the DNA – you can directly add the diluted sample to the PCR. This saves time and effort! If you have particularly valuable sample material, you can of course also use purified samples.
