How does it work?
Shown is exemplary genotyping data from two different blood samples.
The green melting curve originates from an EDTA blood sample and shows a single melting peak at 66 °C. This clearly indicates the presence of a homozygous wild-type genotype.
In contrast, the blue melting curve from another EDTA blood sample displays two distinct melting peaks—one at the same temperature as the green wild-type sample (66 °C), and a second at a significantly lower temperature (54 °C). The presence of two peaks is characteristic of a heterozygous genotype, indicating that two different alleles are present in the sample.
A single melting peak at the lower temperature would, in turn, indicate the presence of a homozygous mutant genotype.
Please note: minor shifts in melting temperatures can occur due to variations in salt concentration and pH in crude samples, which may influence the melting behavior slightly.
