Contents of 1 pouch dissolved in deionized water and made up to 1000mL yields: 1M Tris-HCl buffer, pH7.4 at 25°C.
Sterilization can be performed by filtration. Filtrate the buffer solution through a 0.22µm filter into a sterile flask. Keep the buffer solution at +2°C to +8°C. The pH value of a Tris buffer strongly depends on the temperature. The pKa of 8.06 changes approximately 0.03 units per degree Celsius.
