β-Agarase is used for the recovery of pure DNA and RNA from agarose low melting gels (Ishumatsu K. et al. (1956) Sci. and Ind. (Japan), 30, 137). One β-Agarase unit will solubilise 100mg of low melting agarose gel (1%) at 40-42°C within 30 minutes. To isolate nucleic acids from the agarose gel, cut out the band from the gel as accurately as possible, add to the slice half of the volume (or weight) of 1X reaction buffer and melt the agarose at 40-42°C. Add one unit β-Agarase per 100µL melted agarose and allow to incubate at 40-42°C for one hour. Nucleic acids can then be recovered by a simple ethanol precipitation.
