Penicillin-Streptomycin 10000 U/mL: Penicillin G aus Penicillium notatum und Streptomycin aus Streptomyces griseus verhindern mit ihrer kombinierten Wirkung (Pen-Strep) gegen grampositive und gramnegative Bakterien eine bakterielle Kontamination von Zellkulturen. Penicillin G wirkt einerseits direkt, indem es den Neuaufbau der bakteriellen Zellwand stört, sowie indirekt, indem es die Freisetzung von Enzymen auslöst, die die Zellwand weiter verändern. Streptomycin wirkt, indem es sich an die 30S-Untereinheit des bakteriellen Ribosoms anlagert, was zu einer Hemmung der Protein-Synthese und zum anschließenden Absterben der Bakterien führt.

Penicillin G Kaliumsalz ist in Wasser gut löslich. Bei +4°C oder Raumtemperatur ist die Trockensubstanz ca. 5 Jahre stabil. Die gepufferte Lösung (pH4,5-7) verliert bei +4°C nach 20 Tagen 5% an Aktivität und nach 60 Tagen ca. 20%. Bei Raumtemperatur ist bereits nach 48 Stunden ein 5%iger, nach 7 Tagen ein 10%iger und nach 14 Tagen ein 20 - 50%iger Aktivitätsverlust zu verzeichnen. In einer ungepufferten Lösung ist die Stabilität erheblich geringer (+4°C ca. 3-7 Tage).
Pen-Strep: Penicillin G und Streptomycin gegen bakterielle Kontamination von Zellkulturen.

Change in colour of the Pen/Strep solution:
Normally this change in colour does not mean that you can not use the solution anymore. There are different reasons for it.
1. Chemical reaction of Penicillin and oxygen which will form a substance that absorbs light to get a pale yellow colour. But this does not mean that the majority of the Penicillin is degraded. THere is still enough antibiotic in the solution.
2. The other point is, sometimes, that the Streptomycin source does still contain some pigments from the strain from which Streptomycin is isolated. These pigments may also undergo a reaction with oxygen that turns them more yellowish.

In any case you feel not good about the yellowish colour you should discard it, at this will be the only possibility to have no uncertainty for explanations of further results.
