Highly selective SNP DNA polymerases: The gold standard for allele-specific PCR testing and genotyping

The world of genomics and molecular biology has taken a breathtaking leap into the future in recent years. In addition to "high fidelity" enzymes, "highly discriminating" enzymes such as the highly selective SNP DNA polymerases from Genaxxon have become the superstars of the laboratory. These enzymes are specifically modified for highly selective PCR tests and mutation detection - they help you when you want to find "the needle in the haystack".

What makes highly selective SNP DNA polymerases unique?
Many of the enzymes on the market today have little or no specific and highly selective discrimination. This is especially problematic when your tests rely on specific primers, such as allele-specific PCR (as-PCR). Highly selective enzymes such as Genaxxon's SNP DNA polymerases are the new gold standard for these applications.

Our SNP DNA polymerase has been modified to distinguish between primers that bind completely to the template and those that bind only partially to the template. Or to put it more simply: SNP DNA polymerases distinguish highly selectively between mismatches of the primer-template complex and complete bindings. The enzymes only amplify when there is no mismatch. In this way, for example, mutant alleles can be distinguished highly selectively from wild-type alleles - and without any sequencing at all.

What features do Genaxxon's SNP DNA polymerases offer?
Our SNP DNA polymerases offer some excellent features you can rely on:

  • Excellent SNP detection for reliable discrimination of different alleles.
  • Excellent performance, particularly suitable for allele-specific PCR (as-PCR) and also the first choice for highly selective PCRs.
  • Guaranteed simple primer design with standard primers where you only need to change the 3' nucleotide.
  • Guaranteed highly sensitive. So less than 10 copies of a mutation can be detected in >10 000 wild-type copies.
  • No false amplification thanks to the aptamer-based hot-start function.

For which specific applications are the highly selective SNP DNA polymerases suitable?

Application #1: Genotyping
The SNP DNA polymerases can be used in genotyping. Compared to other genotyping methods such as PCR- or NGS-based methods, SNP DNA polymerases offer decisive advantages: they allow a reduction in overall working time and costs* compared to conventional methods and do not require significant protocol changes. This significantly accelerates your research.

Application #2: Cancer mutation detection
SNP DNA polymerases can be used in allele-specific PCR (as-PCR) assays to detect cancer mutations. To get a fast and reliable result, you can then use a qPCR followed by melting curve analysis. This could speed up your workflow compared to a conventional PCR workflow. In the conventional PCR workflow, primers must be designed, PCR tests must be performed, and the DNA fragments must be separated by gel electrophoresis. With SNP DNA polymerases, this workflow could significantly be shortened: you only need to design your primers and then perform qPCR for simultaneous amplification and detection.

Application #3: CRISPR-Cas SNP Quality Control
SNPs (Single Nucleotide Polymorphisms) can provide important information about an individual's genetic make-up and indicate their susceptibility to certain diseases. SNPs inserted by CRISPR technology must undergo specific quality control procedures. Here, too, SNP DNA polymerases can be used specifically - and in a considerably shorter period than with conventional methods, resulting in a time saving, cost reduction and an overall reduction in workflow costs*.

In which form are Genaxxon's SNP DNA polymerases available?
The SNP DNA polymerases are available as enzymes or as master mixtures. The SNP PolTaq DNA polymerase variant has a 5'-3' nuclease activity and can therefore be used with specific primer probes such as Taqman® probes or molecular beacons.
