NGS DNA Purification MagBeads bieten eine SPRI-basierte Chemie, die die Größenauswahl von Nukleinsäuren und die PCR-Aufreinigung für die Vorbereitung von DNA- und/oder RNA-Bibliotheken für die Sequenzierung der nächsten Generation beschleunigt und vereinfacht. Sie ist mit den gängigen Kits zur Vorbereitung von NGS-Bibliotheken kompatibel. Bei der Solid Phase Reversible Immobilization (SPRI)-Technologie werden paramagnetische Beads eingesetzt, um Nukleinsäuren wie DNA und/oder RNA selektiv nach Art und Größe zu binden. Sie müssen somit bei der DNA-Isolierung bzw. RNA-Extraktion nicht zentrifugieren oder aufwändige Filtrationsschritte durchführen. So ermöglicht SPRI sichere, hochleistungsfähige Protokolle für anspruchsvolle molekularbiologische Anwendungen wie qPCR, ddPCR, Sanger-Sequenzierung, NGS und Microarrays. NGS DNA Purification MagBeads sind mit den meisten Kits einsetzbar, die zur Herstellung von NGS-Bibliotheken (DNA und RNA) angeboten werden. Unser Kit ermöglicht den problemlosen Ersatz von Wettbewerberprodukten (Beads), z.B. AMPure XP Beads. In Kombination mit halbautomatischen und/oder automatisierten Gerätelösungen gelingt Ihnen die effiziente Aufreinigung selbst schwierigster Matrizes bei gleichzeitig reduzierter Fehlerrate und hoher Ausbeute. Wieso Reinigung und Qualitätskontrolle von DNA zur Anwendung bei der NGS?Der Reinigungsschritt ist in der Regel notwendig, um unerwünschtes Material zu entfernen, das die Sequenzierung stören könnte und zu unerwünschten Signalen führt. Einige NGS-Plattformen haben sehr strenge Anforderungen an die DNA-Fragmente, die sequenziert werden können. Das Abtrennen jeglicher DNA-Fragmente, die nicht im akzeptierten Größenbereich liegen ist dann für ein gutes Sequenzierergebnis unabdingbar.  Die optimale Bibliotheksgröße wird durch die Sequenzierungsanwendung vorgegeben. Es ist üblich, die Nukleinsäuren mit Hilfe von Magnetkügelchen zu reinigen. Die Qualitätskontrolle ist der letzte Prozess vor der Sequenzierung. Da die anschließenden Experimente zeitaufwändig und teuer sind, sind strenge Qualitätskontrollschritte erforderlich, um sicherzustellen, dass alle Proben für ihre Anwendungen geeignet sind. Die Bestätigung der Qualität und Quantität der DNA erhöht die Zuverlässigkeit der Sequenzierungsdaten! Das Protokoll unseres NGS MagBead DNA-Reinigungskit kann an die Vorgaben Ihrer aktuellen Liquid-Handling-Workstation (z. B. Beckman, Hamilton, Tecan, Caliper, Perkin Elmer, Agilent und Eppendorf) angepasst werden, wobei Ihr aktuelles Protokoll verwendet wird.
