PBS-solution without Ca, Mg and NaHCO3. Phosphate-buffered saline (PBS) is a balanced salt solution that is used for a variety of cell culture applications, such as washing cells before dissociation, transporting cells or tissue samples, diluting cells for counting, and preparing reagents.
PBS is formulated without calcium and magnesium for rinsing chelators from the culture before cell dissociation.

PBS without Ca and Mg ions, also named PBS(-) is mainly used before 1. trypsinization, 2. washing adherent cells before passaging or 3. to remove serum before enzyme treatment.
PBS(-) weakens cell-cell and cell-surface interactions, thus reducing adhesion of the cells.
