PBS-solution without Ca and Mg and NaHCO3. 10-times concentrated. Phosphate-buffered saline (PBS) is a balanced salt solution that is used for a variety of cell culture applications, such as washing cells before dissociation, transporting cells or tissue samples, diluting cells for counting, and preparing reagents. PBS is formulated without calcium and magnesium for rinsing chelators from the culture before cell dissociation.

PBS containing calcium and magnesium ions is also referred to as PBS(+). Calcium and magnesium ions help cells to bind to one another or to adhere more effectively to a surface. Ca²⁺ and Mg²⁺ therefore stabilise the cell network and cell adhesion, respectively.
PBS(+) is mainly used 1. for the initial washing of cells when cell adhesion needs to be preserved, 2. for tissue handling, or 3. in experiments designed to investigate cell adhesion or receptor activity, where the results depend on the Ca²⁺ and/or Mg²⁺ concentration.
PBS(+) enhances cell-cell and cell-surface interactions and may therefore reduce the efficacy of trypsin action.
