How does it work?
Shown is exemplary genotyping data from two different blood samples.
The red melting curve originates from an EDTA blood sample and shows a single melting peak at 66°C. This clearly indicates the presence of a homozygous wild-type genotype.
In contrast, the green melting curve from another EDTA blood sample displays a pronounced melting peak at a significantly lower temperature (55°C). A single melting peak at this lower temperature indicates a homozygous mutant genotype. Conversely, the presence of two peaks in a melting curve (at 66°C and at 55°C) would be characteristic of a heterozygous genotype, thus indicating the presence of two different alleles in the sample.
Please note: minor shifts in melting temperatures can occur due to variations in salt concentration and pH in crude samples, which may influence the melting behavior slightly.
