The improved thermostability ensures robust performance: Challenger Hotstart RT remains inactive at 42 °C and is only activated at higher temperatures. Therefore, a standard reverse transcription reaction with oligo(dT) primers or random hexamer primers at 42 °C is not possible with this enzyme.

The Challenger Hotstart RT is a genetically engineered reverse transcriptase (RT) that transcribes RNA into cDNA. Its key feature is a "hot-start" property, which is achieved through a specific aptamer-inhibitor.
The hot-start mechanism prevents enzyme activity at room temperature. This significantly reduces the formation of non-specific products, leading to higher specificity and yield in cDNA synthesis. The enzyme only becomes fully active at temperatures above 55°C.
The Challenger HotStart RT is excellently suited for cDNA synthesis. Due to its hot-start property, which only allows activity at temperatures above 55°C, it is ideally used with gene-specific primers whose annealing temperature lies in this high range.
No, the Challenger Hotstart RT is not designed for use with oligo(dT) or random hexamer primers. The reason is a temperature incompatibility: these primer types typically require reaction temperatures below 42°C to bind efficiently.
